Introduction of Lipid Peroxidation in Psychiatry

The brain is known to be sensitive to oxidative stress and lipid peroxidation responses. Recent research data suggest that oxidative damage and lipid peroxidation are seen in major depression, bipolar disorder, and schizophrenia. In some cases, the mechanism of action of oxidative stress may be related to increased membrane lipid peroxidation. For example, ethane levels have recently been found to be elevated in schizophrenia, a direct marker of n-3 lipid peroxidation. Preclinical lipid peroxidation studies have helped to investigate whether measures of lipid peroxidation and oxidative stress can be used to determine the risk of developing various psychiatric disorders and whether long-term disease outcomes can be modified by interventions that reduce reactive oxygen species.

Principle

Taking the detection of MDA as an example, lipid peroxidation is measured by the reaction of MDA with thiobarbituric acid (TBA) to form a colorimetric (532 nm)/fluorescent (λex=532/λem=553 nm) product, proportional to the presence of MDA.
